Influence of β-nicotinamide mononucleotide on the decidualization level of human endometrial stromal cells of patients with repeated implantation failure |

Abstract To investigate the influence of β-nicotinamide mononucleotide(β-NMN)on the decidualization level of human endometrial stromal cells of patients with repeated implantation failure. Methods: Endometrial tissues from 3 patients with repeated implantation failure were obtained by endometrial biopsy, and three primary human endometrial stromal cells were isolated from endometrial tissues after enzyme digestion and cultured in vitro. According to whether β-NMN added and the decidualization of the endometrial stromal cells, these cells were divided into experimental group, control group, and blank group. In the experimental group, β-NMN was added during decidualization of endometrial stromal cells. In the control group, β-NMN was not added during decidualization cultured of endometrial stromal cells. In the blank group, the endometrial stromal cells had not been decidualization cultured. The levels of the mRNA expression and the secretion of the decidual markers, such as prolactin(PRL)and human insulin-like growth factor binding protein 1(IGFBP-1)of the cells during cultured were compared among the three groups. Results: The levels of PRL(2.38±0.10 ng/nl)and IGFBP-1(1.89±0.12 ng/nl)of the cells in the experimental group were significantly higher than those(1.19±0.17 ng/nl and 0.99±0.20 ng/nl)of the cells in the control group(P<0.05), and which of the cells in the control group had no significant different from those of the cells in the blank group. The mRNA expression levels of PRL(15.82±12.92)and IGFBP-1(9.86±8.67)in the cells detected by quantitative PCR had increased after decidualization cultured significantly(P<0.05). Conclusion: β-NMN added in the human endometrial stromal cells during decidualization cultured of the patients with repeated implantation failure can improve the decidualization level of these cells.
