Abstract To investigate the expression of homeobox protein B5 (HOXB5) in endometrial cancer tissue, and to study its influences on the biological behavior of the endometrial cancer cells. Methods: The 35 specimens of the endometrial cancer tissue and 35 specimens of the corresponding paracancerous tissue were collected from the patients with endometrial cancer. The expression of HOXB5 in the endometrial cancer tissue and in the paracancerous tissue was detected by qRT-PCR. The endometrial cancer cells Ishikawa in log phase were randomly divided into CK group (cells were normal cultured), si-NC group (cells were transfected with si-NC), si-HOXB5 group (cells were transfected with si-HOXB5), pcDNA group (cells were transfected with pcDNA), and pcDNA-HOXB5 group (cells were transfected with pcDNA-HOXB5). The transfection efficiency of these groups was detected by qRT-PCR. The cell proliferation was detected by CCK-8 assay, and the cell apoptosis was detected by flow cytometry. Transwell assay was applied to detect the cell invasion, and Scratch assay was applied to detect the cell migration. Western blot was applied to detect the expressions of proliferating cell nuclear antigen (PCNA), B-cell lymphoma-2 (Bcl-2), Bcl-2-associated X protein (Bax), and matrix metalloproteinase-9 (MMP-9) protein in the cells. In vivo xenograft experiment was used to detect the situation of tumor growth. Results: The expression of HOXB5 (1.75±0.13) in the endometrial cancer tissues was significantly higher than that (1.00±0.00) in the paracancerous tissues (P<0.05). The OD450 value, the number of invasive cells, the wound healing rate, the protein expressions of PCNA, Bcl-2, and MMP-9 in the Ishikawa cells, and the quality of the transplanted tumor of the nude mice in si-HOXB5 group were significantly lower than those in CK group and in si-NC group. The apoptosis rate and the protein expression of Bax in si-HOXB5 group were significantly higher than those in CK group and in si-NC group. The OD450 value, the number of invasive cells, the wound healing rate, the protein expression rateS of PCNA, Bcl-2, and MMP-9 in the Ishikawa cells, and the quality of the transplanted tumor of the nude mice in pcDNA-HOXB5 group were significantly higher than those in CK group and in pcDNA group, but the apoptosis rate and the protein expression of Bax in pcDNA-HOXB5 group were significantly lower than those in CK group and in pcDNA group (all P<0.05). Conclusion: The expression of HOXB5 in the endometrial cancer tissues is high. The silencing HOXB5 can inhibit the proliferation, the invasion, and the migration of the Ishikawa cells, and which can promote the apoptosis of the Ishikawa cell.
