Abstract To explore the correlation between LPL gene polymorphism of pregnant women with gestational diabetes mellitus (GDM) and their insulin resistance. Methods: 93 pregnant women with GDM who wanted delivered in hospital were selected in study group from January 2019 to April 2020, and 105 healthy pregnant women were selected in control group during the same period. Polymerase chain reaction restriction endonuclease fragment length polymorphism analysis method (PCR RFLP) analysis was used to detect the LPL Hind gene polymorphism in the placental tissue of these women. The levels of fasting blood glucose (FBG) of the women and their offspring, the levels of serum total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL), low density lipoprotein (LDL), and fasting insulin (FINS) of the women, and the values of insulin resistance index (IR) and body mass index (BMI) of the women were statistically analyzed. The correlation between the IR value of the women and their LPL polymorphism was also analyzed. Results: The H+ allele frequency (82.8%) of the women in the study group was significantly higher than that (64.8%) of the women in the control group. There were significant differences in the levels of TC, TG, HDL, LDL, and FINS, and the BMI value between the women with H+H+ genotype and the women without H+H+ genotype (all<0.05). There was a positive correlation between LPL Hind gene polymorphism of the women in the study group and their insulin resistance index. The values of BMI and IR, and the FINS level of the offspring of the women with H+H+ genotypes in the study group were significantly higher than those of the offspring of the women without H +H+ genotypes (P<0.05). Conclusion: LPL gene polymorphism is involved in the insulin resistance of the pregnant women with GDM, which is related to the insulin resistance of the offspring of these women.
