Expression of microRNA-221 and matrix metalloproteinase inhibitor 2 in serum and placenta tissue in women with early-onset severe preeclampsia complicated with fetal growth restriction |

Abstract To investigate the expression and clinical significance of microRNA-221 (miR-221) and matrix metalloproteinase inhibitor 2 (TIMP-2) in serum and placenta tissue of pregnant women with early-onset severe preeclampsia (EOSP) and fetal growth restriction (FGR). Methods: 256 pregnant women with EOSP were selected as the observation objects from September 2018 to October 2020. According to the FGR situation, these women were divided into group A (139 women without FGR) and group B (117 women without FGR). Enzyme-linked immunosorbent assay was used to detect the serum TIMP-2 level of the women in the two groups. qRT-PCR was used to detect the expression of miR-221 in serum and placenta tissue. Immunohistochemical method was used to detect TIMP-2 protein expression in placenta tissue. Pearson method was used to analyze the correlation between the serum miR-221, TIMP-2 expression levels of the women and their newborn weight, and ROC curve was used to analyze the predictive value of miR-221 and TIMP-2 levels for FGR of the women with EOSP. Results: The levels of MiR-221 in serum and placental tissue (1.65±0.23 and 2.32±0.21), the serum TIMP-2 level (38.52±8.36 ng/ml), and the positive expression rate of TIMP-2 protein in placental tissue (82.1%) of the women in group B were significant higher than those of the women in group A (P<0.05). Correlation analysis showed that miR-221 and TIMP-2 expression levels of the women were negatively correlated with their neonatal weight (P<0.05). The area under the curve of the combined levels of serum miR-221 and TIMP-2 for predicting FGR occurrence of the women with EOSP was 0.921, and which’s sensitivity and specificity were 88.9% and 84.2%. Conclusion: The expressions of serum miR-221 and TIMP-2 in the serum and placenta of the pregnant women with EOSP and FGR are high, and which is related to their FGR occurrence. So the serum miR-221 and TIMP-2 levels are expected to be molecular markers for evaluating FGR of the women with EOSP.
