Abstract To investigate the expression levels and the clinical significance of lncRNA SNHG5 and KLF4 in mononuclear cells of peripheral blood of pregnant women with recurrent unexplained spontaneous abortion (URSA). Methods: 40 pregnant women with URSA were selected in group A, 40 pregnant women with normal pregnancy who required induced abortion were selected in group B, and 40 healthy non-pregnant women were selected in group C from April 2018 to December 2019. The expression levels of lncRNA SNHG5 and KLF4 in mononuclear cells of peripheral blood, and the levels of serum TNF-α, IL-4, and IL-12 of the women in the three groups were detected. Multivariate logistic regression model was used to analyzed the related factors of URSA of the women. Results: The expression levels of the mRNA and protein of lncRNA SNHG5 and KLF4 in mononuclear cells of peripheral blood expression, and the levels of serum TNF-α, IL-4, and IL-12 of the women in group C, group B, and group A had increased gradually (all P<0.05). The expression levels of lncRNA SNHG5 and KLF4 in mononuclear cells of peripheral blood of the women were positively correlated with the levels of their serum TNF-α, IL-4, and IL-12 (all P<0.05). The abnormally high expression levels of lncRNA SNHG5 and KLF4 in mononuclear cells of peripheral blood of the women were the correlated factors of their occurrence of URSA (P<0.05). Conclusion: The expression levels of lncRNA SNHG5 and KLF4 in mononuclear cells of peripheral blood of the pregnant women with URSA increase abnormally, and which are the influence factors of URSA.
