The relationships between the serum AQP8 and AQP9 levels and the outcomes of women with PCOS after IVF-ET |

Abstract To investigate the relationship between the levels of serum aquaporin 8 (AQP 8) and AQP 9 of women with polycystic ovary syndrome (PCOS) and their outcomes after in vitro fertilization and embryo transfer (IVF-ET). Methods: The clinical data of 156 infertile women with PCOS after IVF-ET from December 2017 to October 2019 were collected retrospectively. These women were divided into group A (70 women with pregnancy) and group B (86 women without pregnancy) according to the outcomes after IVF-ET. The general data were collected, which included the age, the body mass index, the infertility duration, the values of basic follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E2), progesterone (P), testosterone (T), and prolactin (PRL), dosage and time of gonadotropin (Gn) used. At the same time, the number of obtained eggs, the rate of mature eggs, the rate of fertilization, and the rate of high-quality embryos were recorded. The levels of serum AQP8 and AQP9 were detected by real-time fluorescence quantitative PCR (qRT-PCR). Spearman method was used to analyze the relationship between the serum AQP8 and AQP9 levels of the women with pregnancy after IVF-ET and their T level, and the rates of mature egg, fertilization, and high-quality embryo. Logistic analysis was used to analyze the factors influencing pregnancy of the women with PCOS after IVF-ET. Results: Compared with those of the women in group B, the serum T and AQP8 levels of the women in group A were significant lower, while the serum AQP9 level, and the rates of mature egg, fertilization rate, and high-quality embryo of the women in group A were significant higher (all P<0.05). Spearman analysis showed that the serum AQP8 level of the women with pregnancy after IVF-ET was positively correlated with their T level, and was negatively correlated with the rates of mature eggs and high-quality embryos (P<0.05). The AQP9 level of the women with pregnancy after IVF-ET was negatively correlated with their T level, and was positively correlated with their rates of fertilization and highquality embryos (P<0.05). The levels of AQP8 and T were the independent risk factors of the outcomes of the women with pregnancy after IVF-ET, and the AQP9 level and the high quality embryo rate were the protective factors of outcomes of the women with pregnancy after IVF-ET (all P<0.05). Conclusion: The low expression of the serum AQP8 and high expression of the serum AQP9 of the PCOS women with pregnancy after IVF-ET are closely related to the outcomes of IVF-ET, which suggest it has some significance in clinic.
